Cisapride
Indications/Uses
GI disorders in adult eg, gastroparesis, reflux esophagitis. Severe gastroesophageal reflux in childn.
Dosage/Direction for Use
Adult
Initially 5 mg to be taken 3-4 times daily. Max: 40 mg.
Childn
Initially 0.2 mg/kg 3-4 times daily. Max: 0.8 mg/kg/day or 20 mg/day.
Administration
Should be taken on an empty stomach: Take 15 min before meals & at bedtime.
Contraindications
GI haemorrhage, mechanical obstruction or perforation.
Special Precautions
Arrhythmia, ventricular fibrillation, Torsades de pointes & QT prolongation.
Adverse Reactions
Dizziness, nausea, pharyngitis, chest pain, fatigue, back pain, depression, dehydration, myalgia, abdominal cramps, borborygmi, diarrhoea, rash, pruritus, urticaria, angioedema, bronchospasm.

Drug Interactions
Increased plasma levels if administered w/ azole antifungals, macrolides, HIV PIs, nefazodone. Increased risk of arrhythmias & QT prolongation if administered w/ antiarrhythmics, tricyclic & tetracyclic antidepressants, antipsychotics, astemizole, sparfloxacin. Increases effects of anticoagulants. May increase absorption of cimetidine & ranitidine. Effects antagonised by anticholinergics.
